I am trying to apply a transition to a slide background but nothing is showing as being selected when viewing the SLIDE TRANSITION area. The blue box in transition popup can be selected but not saving or working after save/publish and preview the page.

Please create me a temporary wp-admin access to your site with admin role.. I would like to check the backend, maybe there is a JS error on your site which prevents LayerSlider's admin interface to save the selected transitions.
I've set the transition Middle-Scaling from right to your first slide. You can select any transitions by just clicking them and make them selected (blue color). If you select more transitions, the slider will choose one randomly on each cycle. It is good to know, that the set transition of the slide is happening when you navigate TO that slide (and not navigating FROM that slide).
