I am wondering if there is a way to play slide 1 the first time the site is loaded. Then play slides 2-x and not play slide 1 again. Like set a cookie so slide 1 doesn't load again?

Unfortunately there is no option to achieve this at the moment. You cannot remove certain slides from the slideshow while it's playing.
